Accounting for Rough Bed Friction Factors of Mud Beds as a Result of Biological Activity in Erosion Experiments ArchiMer

The average bed shear stress and bed friction factor of samples with any roughness was derived from the head loss between upstream and downstream of a test section in an erosion tunnel. The method was validated in both hydraulically smooth (plexiglass; Reynolds number less than 25,000) and rough regimes (calibrated particles with known roughness). As a first step toward using this method on natural sediment, this method was tested with experimental mesocosms assembled from field collected materials (sieved sediments; diatoms). Marine invertebrates are capable of synthesizing bioactive compounds, which may be beneficial to human health. The aim of this study was to evaluate the antioxidant, hemolytic, antimicrobial and cytotoxic activities of crude extract (70% EtOH), and dichloromethane (DCM), ethyl acetate (EtOAc), and aqueous (Aq) fractions of the marine zoanthid Palythoa caribaeorum. The phenolic compound contents of the crude extract, DCM, EtOAc and Aq fractions were 12.33, 18.17, 10.53, and 3.18 mg GAE per gram, respectively. Biological activity of Serratia marcescens cytotoxin BJMBR

Serratia marcescens cytotoxin was purified to homogeneity by ion-exchange chromatography on a DEAE Sepharose Fast Flow column, followed by gel filtration chromatography on a Sephadex G100 column. The molecular mass of the cytotoxin was estimated to be about 50 kDa. Some biological properties of the cytotoxin were analyzed and compared with well-characterized toxins, such as VT1, VT2 and CNF from Escherichia coli and hemolysin produced by S. marcescens. The sensitivity of the cell lines CHO, HeLa, HEp-2, Vero, BHK-21, MA 104 and J774 to the cytotoxin was determined by the cell viability assay using neutral red. Here, we present the current state of knowledge on the diversity of Chilean bryophytes (mosses, liverworts, and hornworts), the secondary metabolites present in these plants, and the biological activity of extracts from populations of Chilean species. Chemical composition, antioxidant and biological activity of Ocotea bicolor Vattimo-Gil (LAURACEAE) essential oil BJPS

ABSTRACT The essential oil composition of the Ocotea bicolor, native plant of Brazil, was studied for the first time. The essential oil of the leaf was obtained by hydrodistillation and analyzed by GC/MS. The analytical procedure revealed a predominance of sesquiterpenes, δ-cadinene (7.39%), β-sesquiphellandrene (6.67%), β-elemene (5.41%) and α-cadinol (5,23%). The essential oil was submitted to brine shrimp toxicity evaluation, antioxidant and antibacterial tests. The antioxidant activity by the formation of phosphomolybdenum complex method presented positive results. We want to construct a yeast expression system for thymosin a1 (Ta1) to make the orally administered Ta1 preparation possible. The whole Ta1 DNA fragment was obtained by PCR. After being digested with restriction enzymes, it was cloned into pYES2 vector. Sequencing was performed to identify the recombinant. The sequence of Ta1 in recombinant coincided with the original one reported in Genbank. When pYES2-Ta1 plasmid was transformed into yeast, galactose instead of glucose was used to induce Ta1 expression. Western blot was performed to identify the quality of the expressed Ta1. This study evaluates the biological activity and possible deterrance played by organic crude extracts from six Phaeophyta and two Rhodophyta algae from Southern Caribbean. Bioessays were carried out using concentrations equivalent to those present in algae tissue. Species in general did not show high citotoxic activity. However Dictyota pulchella, showed a delay in the mitotic process for cells stage XVI. Extracts from Dictyota bartayresiana, D. pulchella, Sargassum cymosum and Cladophyllum schnetteri, inhibited S. partitus feeding under laboratory conditions. Nontheless, only extracts from D. bartayresiana and C. schnetteri showed this activity in the field.

Expression, production, and renaturation of a functional single-chain variable antibody fragment (scFv) against human ICAM-1 BJMBR

Intercellular adhesion molecule-1 (ICAM-1) is an important factor in the progression of inflammatory responses in vivo. To develop a new anti-inflammatory drug to block the biological activity of ICAM-1, we produced a monoclonal antibody (Ka=4.19×10−8 M) against human ICAM-1. The anti-ICAM-1 single-chain variable antibody fragment (scFv) was expressed at a high level as inclusion bodies in Escherichia coli. We refolded the scFv (Ka=2.35×10−7 M) by ion-exchange chromatography, dialysis, and dilution. The results showed that column chromatography refolding by high-performance Q Sepharose had remarkable advantages over conventional dilution and dialysis methods. Background: Antithrombin III (ATIII) is a protein that inhibits abnormal blood clots (or coagulation) by breaking down thrombin and factor Xa. ATIII helps to keep a healthy balance between hemorrhage and coagulation. The present work demonstrated the production, purification and characterization of recombinant human antithrombin (rhAT) from yeast Saccharomyces cerevisiae BY4741 was demonstrated. After expression of rhAT by S. cerevisiae, the biomass and rhAT concentration were analyzed through fed-batch fermentation process.
